In this review, the research progress of 1,4-butanediol (1,4-BDO) production via metabolic engineering was summarized in detail. It was aimed to provide a thorough insight into bio-based production of 1,4-BDO, including the modification of metabolic pathways, the regulation of rate-limiting steps, the control of central carbon flux and the utilization of different carbon sources to produce 1,4-BDO.
